Reactive oxygen species detection method in blood vessels and vascular cells is essential in research studies of cardiovascular diseases. Several fluorescent dyes can be used to quantify cellular ROS, for example dihydroethidium (DHE). DHE can freely permeate cell membrane. Once internalized it is oxidized by cellular O2- and intercalates into DNA. DHE stains chromatin of living cells (excitation 535 nm, emission 610 nm). The fluorescent intensity of DHE can be analysed by a spectrofluorimeter or fluorescent microscopy.
